I use a Lowepro Omni Trekker and really like it. It has been great for travelling with all my gear. I carried it on several trips in the last couple of months and it does a great job of protecting the camera and lenses.
Canon 300D Digital Rebel
Canon EF 100-400mm f/4.5-5.6L IS USM and a Canon EF 70-200mm f/4 L USM
along with a Canon 550EX Speedlite flash and several other lenses (3 more), filters, and all the general stuff we all carry or want to carry.
Review #2
I have an like the Omni Trekker, which is kind of like a laptop bag. It's not really deep enough for the 100-400mm with the tripod foot - the bag will bulge, so I just loosen the tripod foot and twist it a bit, and then it's perfect. It holds too much to comfortably carry for any length of time. This is an old picture. I normally have it packed with the 20D, 100-400, 400 prime, Canon EF 17-40mm f/4 L USM, 105 Macro, 50mm, 28-75, portable drive, Rocket blower, hoods, filters, doodads & thingamabobs, batteries, etc. It's generally packed a bit more than this pic.
Thanks to Scott for photo & comments.

*unique removable/hideaway backpack harness
*front and top zippers for access to gear even when the Omni Trekker bag is in the hard case
*removable tripod straps
*includes rugged metal hardware, a reverse-open lid, laminated mesh pockets, a padded, contoured shoulder strap and fully customizable interior
My Take on the bag:
I find this bag/brief to be very suitable for my purposes, one bag carries all. Keep in mind that if filled completely, it is alot of weight to haul around for an entire day. Even though the hidden backpack straps are well designed, the bag is not proportioned as a normal backpack is, therefore you are limited in shoulder movement while it is strapped on your back. As an example, I used this for one solid day 6AM to 12AM throughout Washington DC. The temperature was in the high 90°(F) and by the end of a long day of walking my back was sore. I have used it in this capacity several times on shorter duration trips with no problems. Overall an excellent bag with many pockets, zippers, and fully customizable! While wearing as a briefcase, the top zipper allows for easy removal of a 1, or 2 bodies with lenes attached.(dependant on the size of the lens of course : )

A view of the top panel that opens up for removal of components. There is also a zippered pocket on the reverse side in which you can keep many things such as CF cards, cloths, spare battery, generally things I want quick access to.
